This integrity relies on faithful duplication of DNA, which is catalyzed by DNA polymerases and their accessory factors. These proteins are involved in numerous interactions to ensure correct nucleotide incorporation and facilitate DNA damage tolerance and repair processes. This dissertation is focused on the structural dynamics and interactions of DNA polymerases and accessory factors related to DNA damage tolerance. Förster resonance energy transfer (FRET) and hydrogen exchange mass spectrometry (HXMS) provided key insights into the dynamics of DNA replication. Using FRET, the interactions between the replicative DNA polymerase and DNA damage tolerance proteins was investigated and the results revealed how a small DNA polymerase manager protein can disrupt the interaction between two essential subunits of the replisome. HXMS was used to probe both the intrinsic structural dynamics of DNA polymerases but also to investigate substrate-induced structural changes in Y-family DNA polymerases. In conclusion, DNA polymerases share dynamic properties related to their functions. The findings described here could be used to alter the structural dynamics of specific DNA polymerases or accessory factors in an attempt to modulate their activity.
